If I disable this:

HKEY_LOCAL_MACHINE\SOFTWARE\Microsoft\Windows\CurrentVersion\OptimalLayout\EnableAutoLayout

Will it only disable the attempt to auto optimize the volume with defrag.exe or will it also disable the layout.ini?

I don't know. There is no official Microsoft documentation for this registry setting, so any information you may find on the internet is guesswork.

I assume it will disable layout.ini. So I keep the Windows Defrag-Tools deleted...

If you want to disable auto optimization, use this:
[HKEY_LOCAL_MACHINE\SOFTWARE\Microsoft\Dfrg\BootOptimizeFunction]
"Enable"="N"

Layout.ini is still being updated, but no files are moved. Works for XP and Vista.

I tested it on Windows XP SP3 and found that "defrag.exe" will already be run when executing the ProcessIdleTasks advapi32.dll call. Sad
its true buddy... m not happy with that

You could try to run a script using auto hotkey and name the exe defrag.exe.
